BGSTAR is a test kit for the determination of (1→3)-β-D-glucan. This information shows the application of this kit on a microplate reader. Read the package insert first for details of the regents.


BGSTAR can be used on a microplate reader by the end point method or the kinetic method.In the endpoint method, higher sensitivity can be obtained as the released pnitroaniline is coupled with the diazo reagent and the absorptivity is enhanced. In the kinetic method, an operator can leave after starting the reaction and the measurable range is wider.

 

 

1. Endpoint method

   

    Calculation

   

Concentration of (1→3)-β-D-glucan in a
sample is calculated from the concentration of standard curdlan by the following formula.

Where AStd is the absorbance of standard curdlan. ABlank is the absorbance of β-glucan-free water. ASample is the absorbance of the sample.Cstd is the concentration of standard curdlan, and Csample is the concentration of (1→3)-β-D-glucan(as curdlan) in the sample.

2. Kinetic method

   

    Calculation

 

Concentration of (1→3)-β-D-glucan in a sample is calculated from the concentration of standard curdlan by the follows.

 

Make a calibration curve.

 x axis : logarithmic concentration of stardard
          curdlan.

 y axis : logarithmic onset time.

 

The formula of the calibration curve:log(onset time) = A + B × log(curdlan concentration) where A and B are constants.

 

The concentration of (1→3)-β-D-glucan (as curdlan) in the sample is obtained with the onset time of the sample on the graph.
