ES cell lines can be established nondestructively from single blastomeres, and still retain their pluripotency. The inhibition of endogenous differentiation and activation of proliferation in preimplantation embryos is helpful in establishing ESC lines.
Studies have demonstrated that media containing 2i (PD0325901 and CHIR99021) dramatically increases the efficiency of
establishing mESCs(Ref.1). ACTH 1-24, adrenocorticotropic hormone fragments 1-24 also improved the survival/
proliferation of mESCs, and enhanced the derivation of mESCs from single blastomeres(Ref.2). When used together,
ACTH2i synergistically enhanced the derivation of mESC lines from ICR and C57BL/6J 0.5-4.5 days postcoitum denuded
intact zygotes, embryos, or isolated blastomeres from 2- to 8-cell embryos when cultured in small volume droplets in KSR
ESC medium with feeders. This method is easy to operate and highly efficient(Ref.3).

Repro-nSC is a combination of two small molecules: a GSK-3ß inhibitor CHIR99021, and a TGF-ß receptor inhibitor SB431542. A study has shown the efficient conversion of hESCs into homogenous primitive neuroepithelium within 7 days when under
synergistic inhibition of GSK-3ß and TGF-ß pathways by Repro-nSC alone or in combination with inhibition of the
Notch signaling pathway (e.g. by RO4929097). These primitive neural stem cells can stably self-renew in the presence of
LIF and 1x Repro-nSC, retaining high neurogenic potential and responsiveness to instructive neural patterning cues
towards midbrain and hindbrain neuronal subtypes, in addition to exhibiting in vivo integration.

Repro-ThiPSC (i.e. SMC4 in reference 1) is a cocktail of four small molecules of specific signaling pathway inhibitors. It can be added directly in media to increase reprogramming efficiency of somatic cells to iPSCs, and to improve expansion and
maintenance of hiPSCs under both with feeder cells and feeder-free conditions. It can be used in combination with any
of the widely used reprogramming gene expression systems (e.g. retro/lenti-viral, episomal, mRNA, or proteins) to
enhance the reprogramming somatic cells to iPS cells. This process is robust, efficient and can be easily scaled-up(Ref.1-3).
